Two questions:
Do I have to fertilize my crop everyday before it's harvested? Or can I fertilize it once and be done with it until I harvest it? Is this the same for reharvestable crops or should I fertilize them again after harvesting them once?
Also with the villagers, I know you can increase their friendship by talking to them everyday, but will talking to them twice increase their friendship more? IE they say something the first time, and then say the same thing for the rest of the day, or does your friendship with them raise just the first time you talk to them that day?

1) you can just fertilize them on the first day and not do it everyday the more fertilizer you use the better they are but no you don't have to fertilize everyday unless you really want to. For the ones that regrow no you do not have to re-fertilize them.
2) The friendship points only go up the first time you talk to them talking to them more than that wont do a thing unless you gift them.

About the fertilizer - you never have to fertilize at all if you don't want to, and really at the beginning of the game (before you get the last windmill which will grind seeds for you) it's really only worthwhile to fertilize once and be done until harvest.
But, once you get that last windmill (the one down by Lloyd's cabin), then you will want to fertilize every day (except when the crop is pick-able) in order to raise the star-level of the crop, then take your highest-star crop and put it in the windmill to get seeds back with the same star-level that the crop was, and you use those seeds to continue the cycle of fertilizing it every day. This is how you eventually get to 5-star crops.
Each 1 crop in the windmill gets you 2 seeds.
Re-harvestable crops like corn and tomatoes and yams, etc, will take longer to raise star-levels than single-harvest crops like turnips, but don't give up! You can fertilize them every single day (even on harvest day, after you've taken the harvestable crop off the plant) and eventually the star level will rise. Making your own fertilizer is better than Raul's and will raise re-harvestable crops slightly faster.
Regarding trees - Yes you can fertilize them too just like regrowable crops and eventually those will reach 5-star fruits as well. Even after they reach 5 stars, I recommend continuing to fertilize, since it seems to affect whether you get 1, 2, or 3 fruits per harvest (fertilizer = 3 fruits, no fertilizer usually means less).
