So... what are the exact requirements to win the fishing contest? Does "Freshness" matter?

The Ducks have given me a large with 9 1/2 quality, 8 1/2 size and it's at 9 freshness... will this win?

It should. It's part size, part freshness. If you have a pretty fresh large fish, it should be a pretty good contender.

You can enter a medium fish as well, but you most likely won't win with one of those. You need a large fish to win.
Your fish has to have 7.5 freshness points or above as well.
Since your fish meets both requirements, I believe you can win ^^

Size is the most important factor in winning. It should be reasonably fresh but the largest fish will win. Quality doesn't seem to matter at all.

An A or S rank large fish will win.

You can always save before you land on Meadow Island and try your fish. If that fish doesn't win, reload and go to Meadow Island and fish as long as you want to. I always managed to land the fish I entered on the day of the contest. Keep your entry in a slot though, If you accidentally sort it into the rest it may become lower due to averaging.

The more you fish on that day, the more fishing degree points you will gain and the larger a fish you will be capable of catching.

You should catch the fish on the day on the contest so that the freshness doesn't have a chance to run out.

Or place them in a red refrigerator, the one that keeps freshness forever. Or even in the machine that raises the rank.

The quality improver would take a few seasons to a year to raise the rank of a A or B rank fish to an A or S rank fish.

It will maintain the rank and freshness of the item but it will not improve it much unless you put it in a several seasons before the contest.

It is much easier to fish until you get a fish which has 8-10 size bubbles filled. A fish that big will win the contest.

I thought it was only the size that matters. :? Now I don't knows if my 9 size large fish will win and I'm don't have enough fish degree to reels in a S rank medium or a A rank large fish yet. I hope my ducks will begins me a fresher 9 size large fish or a 10 size medium fish prior to the contest.

It does have to have at least 7.5 freshness points so don't bring an old fish.

If you are using ducks, go first thing in the morning. Save the night before. If you don't get a good fish, reload and repeat the reload until you have a good candidate. That was the way I got fish fossil and bottles when I was making Fish Natto for my Black Books to finish my shipping list.
